This Ab reacts with the rod domain of human cytokeratin 19 (CK19), a polypeptide of 40 kDa. CK19 is expressed in sweat gland, mammary gland ductal and secretory cells, bile ducts, gastrointestinal tract, bladder urothelium, oral epithelia, esophagus, and ectocervical epithelium. Anti-CK19 reacts with a wide variety of epithelial malignancies including adenocarcinomas of the colon, stomach, pancreas, biliary tract, liver, and breast. Perhaps the most useful application is the identification of thyroid carcinoma of the papillary type, although 50%-60% of follicular carcinomas are also labeled. The mucins are a family of highly glycosylated, secreted proteins with a basic structure consisting of a variable number of tandem repeats (VNTRs). Membrane-associated and secretory Mucins are high molecular weight glycoproteins of the glycocalyx (polysaccharide biofilm) that protects mucosal epithelium from particulate matter and microorganisms. Epithelial Mucins are large, secreted and cell surface glycoproteins crucial for adhesion modulation, signaling and epithelial cell protection. The number of repeats is highly polymorphic and varies among different alleles. The Mucin family consists of Mucins 1-4, Mucin 5 (AC and B), Mucins 6-8, Mucins 11-13 and Mucins 15-17. Recognizes a protein of 130 kDa, identified as Desmoglein-3 (DSG3). This MAb is highly specific to Desmoglein-3 and does not cross-react with other members of the Desmoglein-family. DSG3 is a calcium-binding transmembrane glycoprotein component of desmosomes in vertebrate epithelial cells. Research has shown that DSG3 has a very high sensitivity (80%) and specificity (100%) in recognizing squamous cell carcinoma (SqCC). Therefore, DSG3 is considered a very important marker for lung SqCC and can be a useful ancillary marker to separate SqCC from other subtypes of lung cancer. Spectrin is an actin crosslinking and molecular scaffold protein that links the plasma membrane to the actin cytoskeleton, and functions in the determination of cell shape, arrangement of transmembrane proteins, and organization of organelles. Spectrins function as membrane organizers and stabilizers, composed of non-homologous α andβ chains, which aggregate side-to-side in an anti-parallel fashion to form dimers, tetramers and higher polymers. Spectrin α I and spectrinβ I are present in erythrocytes, whereas spectrin α II (also designated fodrin α) and spectrinβ II (also designated fodrinβ) are present in other somatic cells. BarX1 is a member of the Bar subclass of homeobox transcription factors. Studies of the Mouse and chick homolog suggest the encoded protein may play a role in developing teeth and craniofacial mesenchyme of neural crest origin. The protein may also be associated with differentiation of stomach epithelia. Down-regulation of Barx1 promotes hepatocellular carcinoma cell (HCC) migration, invasion and metastasis, whereas up-regulation of Barx1 inhibits HCC migration, invasion and metastasis. This gene encodes a protein that binds to the hepatocyte growth factor receptor to regulate cell growth, cell motility and morphogenesis in numerous cell and tissue types. Alternative splicing results in multiple transcript variants, at least one of which encodes a preproprotein that is proteolytically processed to generate alpha and beta chains, which form the mature heterodimer. This protein is secreted by mesenchymal cells and acts as a multi-functional cytokine on cells of mainly epithelial origin. This protein also plays a role in angiogenesis, tumorogenesis, and tissue regeneration. Although the encoded protein is a member of the peptidase S1 family of serine proteases, it lacks peptidase activity. Mutations in this gene are associated with nonsyndromic hearing loss. This MAb recognizes a 60 kDa antigen associated with the mitochondria in human cells. It can be used to stain mitochondria in cell or tissue preparations and can be used as a mitochondrial marker in subcellular fractions. It produces a spaghetti-like pattern in normal and malignant cells. This antibody is an excellent marker for human cells in xenographic model research. It reacts specifically with human cells, including neurons and embryonic stem cells. Immunostaining pattern with anti-mitochondrial MAb has been reported as a useful discriminatory adjunct in the complex differential diagnosis of granular renal cell tumors. Recognizes an oncofetal antigen of 220 kDa, identified as a tumor-associated glycoprotein (TAG-72) with properties of a mucin. This MAb defines the mucin-carried sialylated-Tn epitope. TAG-72 is usually expressed by adenocarcinomas, but is negative in mesotheliomas. Studies have reported that this antibody has 80% sensitivity and 93% specificity for pulmonary adenocarcinoma. Therefore, TAG-72 is a useful marker to distinguish between mesothelioma and adenocarcinoma. However, false positive reactions can occur so results must be interpreted with the utmost caution. This antibody may be useful in the differentiation of non-small cell carcinomas from small cell carcinomas of the lung. The combined use of anti-TAG-72 and anti-GCDFP-15 is valuable in the diagnosis of apocrine carcinoma. Recognizes an 85-115 kDa protein (variation with cell type), identified as intercellular adhesion molecule (ICAM-1). It has 7 potential N-linked glycosylation sites. ICAM-1 is a single chain glycoprotein of Ig supergene family, present on unstimulated endothelial cells (EC) and on a variety of other cell types including activated fibroblasts, EC, macrophages, and lymphocytes. ICAM-1 mediates cell adhesion by binding to integrins CD11a/CD18 (leukocyte adhesion molecule, LFA-1) and to CD11b/CD18 (Mac-1). This interaction enhances antigen-specific T-cell activation. ICAM-1 also binds to CD43 and to Plasmodium falciparum infected RBCs. ICAM-1 may also be related to progression and metastasis of tumors. RAD51 is one of the key factors of DNA repair by homologous recombination and has been shown to have anti-apoptotic activity in tumor cells. RAD51 protein interacts with a variety of tumor suppressor proteins including p53, BRCA1 and BRCA2. Elevated expression of RAD51 enhances radio-resistance of human tumor cells. Overexpression of RAD51 protein in tumor cells renders them resistant against cytotoxic drugs like Cisplatin. RAD51 interacts with BRCA1 and BRCA2 to influence subcellular localization and cellular response to DNA damage.
